چکیده
Objective: The study is focused on the SNP in the extracellular domain of TRAIL receptor DR4 at nucleotide 626 in ovarian cancer patients diagnosed in local population in Pakistan. Methods: 16 ovarian cancer patients and 23 control subjects participated in this study. 5ml venous blood was taken from participants with informed consent. DNA was extracted using standard organic methods. PCR-RFLP analysis was done for 626 C→G polymorphism in DR4 gene using site specific primers and restriction enzyme. The results were statistically evaluated in SPSS14. Results: 16 ovarian cancer and 23 control samples were genotyped for C→G polymorphism in DR4 gene. The GG genotype was significantly lower in ovarian cancer patients and controls than the CC and CG in cancer patients and controls. Conclusion: A detailed analysis in future studies and different cancers and cancer subtypes will provide a better understanding of the impact of SNPs in DR4 gene on its transcriptional status.

Tumor Necrosis Factor Alpha -308 G/A Single Nucleotide Polymorphism and Risk of Sperm Abnormalities in Iranian Males
Objective Signaling molecules such as cytokines regulate spermatogenesis during the maturation of germ cells and apoptosis. Tumor necrosis factor-α (TNF-α) is one of the most-documented cytokines that supports spermatogenesis. We investigated the association of TNFα -308 Single Nucleotide Polymorphism with sperm abnormalities in Iranian population. MaterialsAndMethods This case control study in...

Background and Objectives: Genetic modifications in the adiponectin receptor 2 (AdipoR2) gene can affect phenotypes associated with insulin resistance and diabetes. The purpose of this study was to evaluate the possible role of genetic modifications in the AdipoR2 gene, to determine the frequency of genotypes and polymorphism alleles of this gene at rs11061971 (+219 A>T), and to investigate its...

Tumor necrosis factor-related apoptosis-inducing ligand (TRAIL), a member of TNF family, is an interesting ligand which selectively induces apoptosis in tumor cells and, therefore, it has been developed for cancer therapy. This ligand has been produced by various hosts such as E.coli. However, protein expression in E.coli cytoplasm leads to problems such as incorrect folding, reduction in biolo...

متن کاملThe Role of Matrix Metalloproteinase-3 Functional 5A/6A Promoter Polymorphism in Tumor Cell Progression and Metastasis of Breast Cancer
In the human genome, chromosome 11 contains a cluster of matrix metalloproteinase (MMP) genes. Single nucleotide polymorphisms in the promoter region of MMP genes are important for MMP expression. A common adenine deletion polymorphism (5A) at position -1171 of the MMP-3 gene promoter (5´-AAAAAACCAT-3´ change to 5´-AAAAACCAT-3´) facilitates transcriptional factor binding and MMP-3 promoter acti...
